In Vivo Genome Editing: Targeted Delivery & Large Gene Knock-ins


CRISPR gene editing has the potential to redefine treatment of genetic diseases. In order to reach its full potential, two remaining problems need to be solved: targeted delivery and large gene knock-ins. Targeted delivery means getting the CRISPR machinery very specifically to the organs and cell types of the body that will yield a change to the phenotypic expressions of the disease. Many diseases, however, can be caused by hundreds of different mutations, which would require a large number of different therapeutics for each disease. Once we gain the ability to knock out a mutated exon or gene, and knock in a healthy version, all genetic variants of a disease could, in principle, be treated with a single therapeutic. This workshop will address targeted delivery and large gene knock-ins from the perspective of the current state of the technologies, the clinical trials, and the companies and academic institutions working in the space.
